ABSTRACT

Arecae Semen, as the first place among "Four South Medicines" in China, has great dual-use value of medicine and food. The research of Arecae Semen was mainly focused on the active ingredients and efficacy value, and its potential safety hazards were also concerned. Until now, there is still a lack of clear boundaries between medicine and food, resulting in its safety cannot be guaranteed. Therefore, it is of great significance to establish clear boundaries of medicine and food use and health risk assessment. In this paper, the differences of pretreatment and application methods of Arecae Semen were analyzed, and the research progress of Arecae Semen in chemical composition identification and toxicology research and safety evaluation were reviewed emphatically. Finally, the differences of quality control and safety evaluation of Arecae Semen in pharmacopoeias or standards were analyzed at home and abroad. It was expected to provide reference value for quality control, safety evaluation and international standardization research of Arecae Semen.

ABSTRACT

@#Corneal disease is a common disease that causes blindness, and keratoplasty is the main method for the treatment of corneal blindness. Visual function examination alone cannot fully evaluate the impact of surgery on patients, while the vision-related quality of life scale covers all aspects of visual indicators, which can more comprehensively reflect the feelings of patients. In recent years, it has gradually become a common method to evaluate the therapeutic effect of keratoplasty. This paper briefly reviews the research progress of NEI-VFQ-25 scale in corneal transplantation in recent years.

ABSTRACT

Uranium [U(Ⅵ)] in the blood is known to form stable complexes with apotransferrin (apo-Tf), which plays an important role in mediating the cytotoxicity induced by U(Ⅵ) transported to cells. The present study aimed to establish an new in vitro screening model of U(Ⅵ) decorporation agents through exploring the capability of chelating agents competing with U(Ⅵ) binding to apo-transferrin based on enzyme-linked immunosorbent assay (ELISA). The optimal concentrations of apo-Tf coated antigen, Tf antibody, secondary antibody and U(Ⅵ) treatment were achieved and the stability and reproducibility of this method were validated by methodology study. Using this model, the ability of four chelating agents to mobilize the U(Ⅵ) binding to apo-Tf was evaluated, and the rank of competitiveness was catechol-3,6-bis(methyleiminodiacetic acid) (CBMIDA) ≈ Tiron > apo-Tf > DTPA-CaNa3 ≈ DTPA-ZnNa3. The efficacy of these chelating agents in removal of U(Ⅵ) was tested by animal experiments. The results showed that immediate administration of CBMIDA or Tiron after injection of U(Ⅵ) in mice significantly promoted urinary U(Ⅵ) excretion and reduced U(Ⅵ) accumulation in kidneys and femurs, while DTPA-CaNa3 and DTPA-ZnNa3 have no obvious effects as compared to U(Ⅵ)-exposed mice alone, which was consistent with the results of competitive ELISA method. The animal experiments conform to the rules of the Animal Research Ethics Committee of School of Pharmacy of Fudan University. These results show that the new proposed method is rapid, simple and convenient with good reproducibility and has the potential to be used for in vitro screening of U(Ⅵ) decorporation agents.

ABSTRACT

Objective: To investigate the changes in the content of notoginsenoside R1 (R1), ginsenoside Rg1(Rg1) and amino acid neurotransmitters in brain micro dialysate after the treatment with Panax notoginseng saponins (PNS) solution in Parkinson’s disease (PD) model mice induced by 1-methylmethyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP). Methods: Mice were injected intraperitoneally with MPTP to induce PD mice model. After intravenous injection of PNS solution, brain micro dialysate was collected by microdialysis technique. The sample was divided into two parts, one was subjected to pharmacokinetic study of R1 and Rg1 by UPLC-MS/MS, and the other part for pharmacodynamics study which was done by pre-column derivatization of the amino acids and HPLC-FLD analysis. Results: After iv administration of PNS to the mice, the maximum CSF concentration was (35.48 ± 24.4) ng/mL for R1 and (75.61 ± 41.35) ng/mL for Rg1. The time to reach maximum concentration was found 1.75 h both for R1 and Rg1. The AUC value of R1 and Rg1 were (100.12 ± 84.29) ng∙h/mL and (218.84 ± 144.73) ng∙h/mL, respectively. Moreover, the levels of aspartic acid and glutamic acid were reduced, and glycine, taurine and γ-aminobutyric acid levels were increased in PD model mice treated with PNS in comparison with model group. Conclusion: PNS can enter the brain through the blood brain barrier (BBB) to some extent, and regulate the imbalance of amino acid level in PD model mice, so as to display the neuroprotective effect and its therapeutic value.

ABSTRACT

Objective:To study the perceived organizational support,organizational approval impact on job engagement process and influence mechanism of non-establishment staff in public hospital under the background of China's personnel sys-tem,and provide reference for hospital managers to take measures to promote the staff's job involvement. Methods:650 non-es-tablishment staffs from three tertiary public hospitals in each of the six cities of Sichuan province were surveyed by issuing or-ganizational support questionnaires,organization approval scale and work input scale. Descriptive analysis,correlation analy-sis,regression analysis were performed with the help of SPSS 19.0,a three-variable structural equation model was established by using AMOS17.0. Results:The average score of organizational support of non-establishment staffs in public hospitals was (2.80 ±1.05),the organizational approval scale was(3.73 ±1.03)and job engagement was(2.99 ±1.58);and there was a sig-nificant positive correlation (P<0.01) between organizational approval and job engagement. It was also revealed that per-ceived organizational support had a direct effect on job involvement,and indirect effect on organizational approval as a partial mediation variable. Conclusion:Public hospitals have a low level of perceived organizational support,job involvement of non-establishment staffs,and public hospital administrators can promote the job engagement of non-establishment staffs in public hospital by improving the sense of organizational support and organizational identification.
